I have been tasked with creating a style guide for creating in Adobe DC Pro. How do I set the dimensions and position of boxes that will contain task instructions?

You should not be using Acrobat to create files from scratch. Use InDesign, or a similar layout application.

Thanks try67..is it possible to do this though. Can I set dimensions of boxes that will contain instructions. If so, how can I do this? Should I tell my Project Lead that the project is moot?

Not sure what you mean... What kind of "boxes"?
It's definitely doable, but highly not recommended to do it in Acrobat.

Well, like this. I need to be able to indicate the dimensions and location of a box like this, so that someone making the document from scratch will know what size to make the box

You can use the Add Text tool to add text to an existing PDF, but again, it's not a good idea.
You can even create the file in Word and then convert it to a PDF if you don't have InDesign.
Otherwise, hire a professional to do it for you. Just make sure you get both the PDF and the original file format used to create it.

So, say the dimensions of that box is 3.25" long by .25" wide and I want to make a rectangle with those same dimensions on a blank document in Adobe DC Pro, how do I do that?
I just want to know what I need to tell my Project Lead who asked that I create a style guide, which contains the location and dimensions for all fields in the document.

What try67 is saying is correct: do this in a program that is meant to do these kinds of things. While you CAN do this in Acrobat, doing so is more like showing that it can be done as opposed to being the best place to do it.
Word can do this (but is not very good at it) and either InDesign or Adobe Illustrator are perfect for this. Once it's created than convert that document into a PDF and you're good. No great because you didn't waste your time in Acrobat.
But if you really really really want to do this in Acrobat, here's one way to do this: In Word create a new document with nothing on it. Now save it as a PDF.
Now open that document into Acrobat, now make sure your rulers are showing (Command/Control-r) and go to the Comments tab. Select the Drawing Tools, select the rectangle toll and draw away.
The problem you will have here is that you are unlikely will be able to place these rectangles in the regions where you want them. That is you have no perspective where the rest of the page's content are likely to be nor how much space they will use.

You can use a text field for that. You can specify its exact size under Properties, Position.
