Preserve Underlying Transparency Checkbox Missing

I searched online and found that the Preserve Underlying Transparency should be next to the composition and represented by a T symbol.
However, I cannot find it for my PNG files. I believe I have alpha channel fully enabled. Any reasons and solutions to this?

First, the Preserve Underlying Transparency will not have any effect on your PNG files. You are misunderstanding it's function. Put a layer at the bottom of the stack that has transparency - like a text layer, turn on the PUT switch on all the layers above and the text layer will act as an alpha channel for all layers. Here's a blue solid with a red solid to cover the right half of the screen and a gray text layer at the bottom providing the matte for the two layers above it.

Second, your timeline is set to view only the Switches column. The PUT switch is in the Modes column because it is a mode. You can cycle between the Switches and Modes columns by Pressing F4 or by clicking on the Toggle Switches/Modes icon visible in your screenshot at the bottom of the timeline. You can also show both the switches and modes columns by turning on the middle icon at the bottom left of the Timeline Column's area as shown in my screenshot. You can also turn on both by using the Columns popup menu by right clicking on any of the column names.

You also have the Channels and Color Management selector in the Composition Panel set to view only the Alpha Channel so you will only see a black and white image. To determine whether or not your layer has transparency you need to turn to the Transparency Grid (first switch left of the comp camera selector at the bottom of the composition panel) or use the info panel and check the color values of the alpha channel by moving the selection tool over the image in the composition panel.

I'm not sure what you are trying to do with the Preserve Underlying Transparency but because of your Channel and Color Management  is set to view the Alpha channel everything that is white should be opaque and everything that is black should be transparent if you turn on the Transparency Grid in your comp panel.

There is an awful lot that goes on in the Comp Panel and the timeline. It might do you some good to do a little studying on the UI.
