I'm working with the newest version of AE (22.0.1 Build 2) and just creating some extremely simple name keys, but upon trying to export the rbg+alpha option is greyed out. Why?
It's a simple Classic 3D comp with two lines of text, no animation, attempting to export Quicktime then open the drop down menu to choose rbg+alpha only to find that it's greyed out. This is exactly the same workflow that I used in the old version so I don't understand why it's suddenly not working. Is it a bug Adobe needs to fix?

Sorry, I should have added that choosing a format other than Quicktime then clicking on the 'Format' drop down menu a second time and going back to choosing Quicktime makes the rbg+aplha option available again.

Most codecs don't support exporting RGB with an Alpha Channel.
The codec shown in your screen shot Apple ProRes 422 is one of the ones that don't support RGB + Alpha.
Try switching it to ProRes 4444 by clicking on the Format Options button

Although this may be an old post, I just wanted to share that this solution resolved my problem of not being able to see the RGB+Alpha option. It's now 2023 and I'm grateful for the help.

There is now, and there always was an Output Module preset that includes an alpha channel. It always works to just select the High Quality with Alpha preset from the dropdown list. If you want to create your own preset, you first need to study up on formats, video standards, and compression so you don't foul things up.
