Is it possible to change the Font Location using Extended Script

Fonts are handled by your system, so I'm not sure why this is even a question. Unless fonts are in specific folders they won't bee seen by apps and that is that. Moving them around arbitrarily is therefore useless and irrelevant.

Yaa, If the fonts are at installed at specific location and I want to point this location using extended script

Again, no such thing simply because the fonts won't be loaded. If at all, you'd still have to create a link/ alias for each file and place those references in the system's font folder, but then again you could just as well move the actual font files...
