In Audition CS6, when I press spacebar or the play button, nothing happens but in the lower left it turns to "playing." The red line wont move. No audio. Nothing. Then when I open the preferences tab, suddenly it starts to work and play sound in the background. Then when I close the preferences tab is suddenly stops working. Wtf is going on?

Make sure that in Preferences/Audio Hardware page that the box 'Release ASIO driver in background' is not ticked. What audio hardware and drivers are you using?

To expand a bit on what ryclark said, if that "release ASIO drivers in background" is ticked, it allows other programmes to grab your sound card and some aren't very good about releasing it when Audition tries to access it.
Besides unticking the box ryclark mentioned, if it still won't work, go to Edit/Preferences/Audio Hardware and check that it shows the proper choice of driver and audio hardware. If that is right, change any one thing and okay it, then set it again to the proper settings.
If this isn't enough, go to Windows Audio and make sure the proper device is selected there. If not, re-select it as the default.

I did wonder about this a bit yesterday as the behaviour, if it's exactly as described, doesn't quite make sense, on the face of it - although what ryclark is suggesting is the most obvious thing to try. If none of this makes any difference, it might be worth downloading ASIO4ALL and seeing if that provides any clues as to what's going on...
