I am creating my Podcast on Audition. My mics are connected to my Behringer XR 12 mixer. If I connect my USB ouptput to my computer, will Audition see it in the Audio Hardware input in the Podcast template? Thank you.

It should show up as a two-channel device, I think. To work as a multichannel input device you'd need an ASIO driver for it, and I'm not sure if there is one, as this isn't its primary use, after all - it's intended for wireless connection to portable devices. You also need the app in place to control it.

The other thing you should note is that it really isn't a good idea to use the podcast template, as it has a load of inappropriate settings built into it that have caused many users a load of grief. Just start with the default one - or if you really feel that you want to use it, then get all of the effects out of all of the tracks before you start.
