Which theme do you use? Question slides is the only area where you cannot use shape buttons, which is sad. The used buttons are normal buttons, most included themes use a Transparent button style without states, probably a legacy bug. It doesn't help to add a rollover/down style in the Object Style Manager, those states do not appear in the question slides. You can change to a Text Button style, but formatting is then limited to text only, you cannot change the fill nor the stroke. The third type of normal button is the image button, which has states. To configure them is cumbersome: you will have to create 3 different images (ending on _over, _up and _down) for each quiz button (because the label needs to be part of the image). Not really the answer you expected, I don't know why the transparent button states do not appear in question slides, they work well in other slides. Whatever button style you choose, you will only have the InBuilt styles which you can define in the Object Style Manager, there is no way to add another state, the State View panel is not available for those buttons.
