I'm using Captivate 2017 and added a Drag and Drop interactivity to a slide. When I go to preview in either project, from this slide or in browser, it does not work. When I preview in HTML5 browser it works. I am not publishing in HTML5, does that mean I cannot use the Drag and Drop interactivity?
That is THE missing information. Drag&Drop objects always have to be present at the beginning of the slide. Have a look at this older article (still valid): Drag&Drop tips - Captivate blog
The previous details were misleading, there is no difference between wizard or panel use, changing or not changing the feedback messages will change the requirement to have all objects from the start of the slide. You are creating a normal or a responsive project? I'm asking this, because responsive projects can only be published to HTML5.

When I publish I check Scalable HTML content, but I only publish in SWF. Does Drag and Drop only work in html 5?

I've noticed the problem. When I go to change the success/failure messages, the drag and drop guide line (that blue line), disappears. It's as if changing the font, color or even the text deletes the drag and drop guide line.

Answers crossed each other.
You mean the line that connects the drag sources with the drop targets? Is the eye icon in the D&D slide OK? All colored lines in D&D disappear if the eye icon has a red cross line. Clicking on it should bring back all colored lines.
BTW I double-checked this in 2017, and preview worked perfectly.
Must be something else. Having the lines visible or not doesn't matter for the Preview, it is just for editing reasons.

No, I also use it for SWF output. Just wanted to exclude that you didn't use a responsive project.
Next question: are you sure it is not a Flash security problem, you are able to see other slides with the exception of the D&D slide? Recently Flash players are enabled by default in some browsers, that could be another cause? But in both cases you wouldn't be able to see any slide, not only the D&D slide. Can you check that (you can hide the D&D slide to test).

It previews fine if I leave the success/failure message as: Type success message here OR Type failure message here. The moment I change the text, the drag and drop arrow (guide) line disappears and when I preview, it doesn't drag and drop.

Yes, the line was disappearing because of the visibility icon turning off. I've fixed that. This is frustrating because when I create two smart objects and click the + symbol under the drop and drop tab, I can create a drag and drop with not preview issues. When I use the wizard, I get the issues on preview.

OK, I never use that wizard, also work directly from the D&D panel.
I double-checked with the wizard. Then I changed the Success and Failure messages, don't see any issue.
Such problem is difficult to resolve, cannot reproduce your issues. Can you restart Cp and try again?

I'm narrowing this down. If I have the drag and drop at the beginning of the slide, it works fine. If the drag and drop comes in towards the end of the slide, the drag and drop is not working. I think it could be a pausing issue. I'll post a result if I can get it to work, otherwise, I'm just going to have the drag and drop open in a new slide at the beginning rather than at the end of the previous slide.

Thanks. I think this thread will be helpful is someone else stumbles on this issue. I think it was a matter of elimination and this is the first time I've used drag and drop.

If you checked out first some articles about the subject, would have been saving time.
Too bad that the official documentation is not always helping.
This has been answered several times on this forum about D&D needing from the start of the slide.
It is just that you did mention things that couldn't have been the cause like disappearing lines, wizard vs panel, editing messages being the cause. That was the reason I didn't ask for a timeline shot, which I normally always do.
