We're wanting to add a custom field besides e-mail and password in the user profiles of Connect, but we'd also like to have AD populate the custom field. Is there a way of doing this?

Yes, you have that option. See this page:

You can add fields using Adobe Connect Central and tie those to LDAP fields. See the install guide around page 32.
