Which software should I download for a start in this case?

Hi, I am learning how to use adobe creative cloud softwares. Say I want to show to my students a photo from a Microscope field. I want them to identify where is a tiny bug. I believe I have to create a 'hot spot' where the bug is that is transparent. If the student succeed and click on it new links may open, like for quizes, etc.

May I ask you which software or combination of them in the creative cloud should I explore?

It sounds like you're trying to build a web site.  Hotspots on images (aka client-side Image Maps) can be created in Dreamweaver with HTML code.

Use Photoshop to create & edit images. 

Use Dreamweaver to create & upload files (HTML, CSS and JS) to your server.
