What's the best way to create a rendered painting in illustrstor?
If you're asking if Illustrator can create an illustration such as you show then the answer is no. This image doesn't look like it has been painted from scratch. Instead filters and effects have been used to make this look from one or several photos. My husband does this kind of things. There is no one-click solution. It still requires a lot image editing, preparing several copies of the same photo to apply several different operations on it.
There are tutorials how different artists approach this type of job. I would suggest you ask in the Photoshop forum since they definitely know a couple of good tutorials and if you need additional tutorials, YouTube should have plenty to try out different approaches, since it also depends on your source material.

Illustrator is only a tool. It is not an automatic creator of art and design. If you want to create something like this and have the skills to draw and/or paint then learning about the tools available in Illustrator so that you can use them in the same way that a painter or illustrator would traditionally do so is the only way to make art and/or design happen.
Seeing your reply To Ton's suggestion about using Photoshop (after I posted): my same sentiments about Illustrator would also apply to Photoshop.
