I saw a video of how to use Generative Recolor on line art, but that line art seems to have been made in Illustrator. Can Generative Recolor be used on hand drawn line art (black ink on paper) that is scanned and imported into illustrator?

Generative Recolor works on vector artwork in Illustrator.

You will need to convert your scan to vector first. You can use Illustrator's Auto Trace command or you could redraw the line art using the Pen tool.
