Poster Settings?

I need to make a poster for a conference. I am told that the maximum dimensions are 36x48 inches. What resolution should I set my photoshop file to? Do I just leave it at the default, 72 pixels/inch?

The rule of thumb for printing is resolution in the 240-300 px/in range, but for a large poster 72 px/in may work for you, as the observer's eye is at a greater distance compared to a small print.

There is nothing to prevent you from increasing the resolution with re-sampling via Image>resize>image size if the print output is sub-optimal for your purpose.
