An alpha channel exists when part of the image is transparent. Does your video already have that transparent area?

You should be able to use the Apple ProRes MXF OP1a Format with Apple ProRes 4444 with Alpha to encode with an Alpha Channel in an MXF container, assuming like John mentioned already that there is transparency in the video.

What happens with that file subsequently would also dictate the alpha. Like, a video player may not support an alpha channel. Typically, at least to my knowledge, you would only use it in post production software.
