New PC with horrible jerky timeline playback

On a very old workstation with dual Intel Xeon E5-2660, 32GB Ram and a GTX1050 I had no problems to edit HD content. 

 

Now I got a new PC:
AMD Ryzen 9 5900X

Asus X570 PG Velocita

64GB Ram 3200-16 Vengeance LPX

MSI 10GB D6X RTX 3080 VENTUS 3X Plus

 

In both cases the system and Premiere are on a SSD and the content on a HDD.

 

On the new PC I can not play back the same content with no effects in full resolution. After a few seconds the playback jerks and will be set a few frames back every second. Audio is affectetd too. What could be my mistake? Renderer is set to GPU(Cuda).
