There is a possibility that you deleted or moved the file you were working with before letting the Premiere Pro project close for the last time, that means you have your clip offline as Ann says. Make sure the file is in the exact same directory you imported it from, or as Peru Bob says, close Premiere Pro and plug in the external drive you've been working with so your clip will automatically reconnect.
