Hi, I have a rec in 59,995 fps and I think it's a problem for premiere pro because when I import it there is a desynchronisation at the end.

Does I have to put all my screen record at 60 fps constant in handbrake before modify it in premiere pro ? Because I have 2 and a half hours of video so the encoding will be sooo long

Short answer: Yes.

Long answer:

Please use the free MediaInfo and post a screenshot the properties of your media in tree view:

Many users are having issues with VFR. If the file is variable frame rate, use Hand brake to convert to constant frame rate:

To be sure, the best quality on handbrake is 0 FM right ? And last question can I select 60 fps with a record which have less than 60 fps ? (59,995)
