I'm building a new PC and have 2 Tesla P4 cards at my disposal. Is this a good GPU for Premiere? Can I use both of them? What would be the best motherboard and CPU to pair with this card if I can iuse it?

Without a P-series Quadro GPU, a Tesla P4 card is useless. This means that to use even one Tesla P4, you will also need a P-series Quadro card, such as a Quadro P4000 or P5000. And it's because a Tesla card has absolutely no video out at all whatsoever (no place at all whatsoever to connect a primary display to).

And to use all three cards (both of your P4s and a compatible Quadro P#### card), you will need a HEDT (based on either an Intel X-series i9 CPU or an AMD Threadripper CPU) to run them all on. A mainstream PC will not utilize all three of those cards to their fullest due to their lack of a sufficient number of PCIe lanes available to the GPUs.
