This is my graphics card Nvidia Quadro RTX A4500 20GB GPU My question is, do I need to render in HEVC to use the card cores? In my second question, when I try to output in .MOV or other formats, the following message appears on the screen. Please help me, I would be very grateful. Your system's hardware does not suppport hardware acceleration for the current settings. This message does not appear in HEVC, you need to exit mov apple to make a transparent video.

I'm sorry to tell you this, but Windows does not natively support the .mov container wrapper. Hence, no Windows version of Premiere Pro or any other NLE supports hardware acceleration at all for that container. You'll need to edit on a Mac for that.

 

Also, the H.264 and HEVC hardware support in Windows is restricted pretty much to the .mp4 container format - and you must choose an H.264 or HEVC preset for that to work.
