When I do this, and try to access the administrator page, I get a 500 error (when I switch back, it works again). What are the minimum permissions I need to grant the service account to get it to work.

Take a look at the lockdown guide: http://www.adobe.com/content/dam/Adobe/en/products/coldfusion/pdfs/cf10/cf10-lockdown-guide.pdf for more information. There are certain folders in the ColdFusion installation location that need to be granted specific read or read/write permissions to whatever user account you are using for the service. Don't run as an administrator, but run as a user with just enough permissions to access what you need it to access. If you are on a Windows domain, use a domain account and make sure any network resources (file shares) have appropriate permissions granted to that domain account.
