I think this is a Spark question, not a Dreamweaver question. And it looks like the Spark uservoice already has a feature request for this with 36 votes.
enable autoplay as an option – Community feature requests for Adobe Spark!
Keep in mind that some browsers don't support autoplay especially if your video has an audio track. Media that automatically plays on page load is widely regarded as intrusive. It should always be the user's choice to play video or not.
