After a Windows 8.1 update this morning, 11/16, Flash 27.0.0.187 no longer works in Opera 49.0.  I tried uninstalling and reinstalling just now with no results, and was also unsuccessful checking browser settings, although I'm little more than a rookie at this and might have missed something.  Can anyone help?

What does "no longer works" mean?  Please be more specific in describing the issue, and include a direct link to the content.

With the release of Opera 49, Opera now disables Flash Player by default and users must enable it per site.  Please do the following:

    • Ensure you have Flash Player PPAPI installed
    • With the Opera 49 release, Opera now disables Flash by default and users must enable it, per site.  This is the same behaviour Chrome implemented several releases ago.  Since Opera is based on Chromium, they are now also implementing this behaviour.  If you're not prompted to allow Flash when first visiting a site containing Flash, you may need to go to Opera's Settings to add the site to the 'Allow Flash' list.  To do this, do the following:
      • Launch Opera
      • Go to Menu > Settings
      • In the Settings page, click Websites
      • In the right panel, scroll down and locate the Flash section
      • Click on the Manage exceptions... button
      • In the Flash exceptions window, add the domain you want to enable Flash on, for example, helpx.adobe.com, ensure the Behaviour is set to Allow:
      • Click Done
      • Restart Opera

Flash should work.  If not, it's either a content issue (content not configured to handle Opera's click-to-play implementation), or a browser issue.
