Is there a tool in Acrobat that can measure ink coverage on individual art files?

You can use the Preflight tool (Edit → Preflight...) to analyze ink coverage. Also there is a Windows application APFill Ink Coverage Calculator to calculate CMYK coverage for PDF files.
