I have set fields as 'required', but they are not prompting me to complete them when I save the document in 'preview' form. Can you please help?

The only way to do that securely is to have them digitally sign it.

That's correct. Required fields are only validated when the form is submitted.
If you want to use them to validate it when the file is saved or printed you can use a script, like this one I've developed:

Also, if you mean that you're using the Preview app on a Mac computer, don't. It's a very buggy application that corrupts PDF forms.

Submitting a form means sending the entire file, or just the form data, using email or a web-service, back to the person who created it.
You can add a button that will allow the user to do it, and it will automatically validate the required fields when pressed. See: Setting action buttons in PDF forms, Adobe Acrobat

I have done all of this yet the form I created with required fields was emailed back to me by a colleague who said she purposely missed a field or two and she received no warning when saving or emailing the form that they had been missed. Is there a facility that I can send my form to for checking please?

It's possible they just attached the file to an email manually and then sent it to you, instead of using the Submit button. There's no way to prevent someone from doing that...

Can I run a simple script or button to lock the form down once it has been submitted, so that when a client has sent me an application I cannot amend it?
