Wie kann man den standardmäßigen Duplexdruck dauerhaft deaktivieren? Häkchen für Duplexdruck erscheint immer wieder beim Öffnen eines neuen Dokumentes.

As per the description above, you want to disable the duplex printing in Adobe Reader, Is that correct?
The Duplex printing feature is based on Printer settings, to disable the duplex printing, please refer to the following KB docs
