Basic question regarding Adobe Podcast recording protocol

For final audio processed through Adobe Podcast, does the transcript matter to the final recording? Should I bother to edit sentences with periods and capital letters etc.? Does it make any difference in the final mastering/finishing?

Finally, what is the best way via Adobe Podcast to insert a short (2-3 second) pause into audio conversations during editing. The equivalent of a space break or drop cap break in print.
