adding Cue markers in a file and saving it as mp3 file. not deleting it before saving. will it affect mp3 while playing it in music player, will it create any disturbance. Is it essential to delete Cue marker before saving and closing the file?

Since you can't save cue markers in an MP3 file (the format doesn't support them), then no, it won't cause any disturbance at all, because they won't be there!

You can only save cue markers to wav files in Audition, and in fact the markers are stored after the audio in a separate file extension. This is why (on Windows machines, but not Macs) you can use CueListTool to edit and manipulate them externally. In theory you could use the same principle on Macs, but the software has never been ported. You might be able to use it in one of the emulators available, though.

But in principle, Cue markers never affect any audio at all.
