Preferences allow a tolerance to be set for anchor selection but what about clicking on a path? I find I have to get too close to a path to select an object - can I vary that?

You can put in a feature request to the Adobe team here:
Illustrator Feature Requests

Also remember that the tip of the mouse is the part that is active. (You may already know this.)

Can you perhaps show us an example of your artwork? Are there too many paths? Do you have a high res monitor?

Have you tried Smart guides?

No, I just mean in general. I like to have 'Object selection by path only' checked. Selecting even something like a simple rectangle would be easier if one could click slightly further from the path. I find I sometimes miss it. Can't see how smart guides help although I do normally have them turned on.
