What operating system are you using? If you are using any version of the Mac OS after Yosemite (10.10) or Windows 10 then CS6 is totally unsupported. As a matter of fact, Windows 10 periodically runs automatic updates for most users with the result that an unsupported version of Illustrator that may have been running OK up until now (by the greatest of miracles) may begin to act erratically or not work at all after a Windows 10 update.

I personally don't know anything about Wacom tablets but I would suggest—purely as a form of troubleshooting—that you uninstall it and see if the problem with Illustrator persists. I would also check with Wacom as to whether the version you installed is compatible with Yosemite and Illustrator CS6. If it is a very recent drive then it is likely that it is configured to work with a much later version of Illustrator and the Mac OS.
