defined brush not greyed out and not working

After creating the selection of less than 2500 by 2500 pixels, selecting edit->define brush preset and giving it a name, I see the new brush in the brush presets, but it is empty and paints nothing.  Opacity is 100 percent.  I've done it before, but it doesn't work now.  Why might that be?

OK, the deal is that brushes are made from selections and what is selected must not be white, but be black.  I'll have to experiment to see how it all really works, but basically "select black colors for your brush".  Meanwhile, I need to delete the useless brushes.  The delete key is too obvious and doesn't work.  I have a number of entries in the "brush presets" now that I have no use for and the way to get rid of them is not especially obvious.

Are any brush presets applied?

What colour were the pixels that you defined the brush from? 

Was the brush defined from an otherwise transparent layer?

I don't know what is meant by "brush presets applied".  I had a selection and said "make a brush from the selection".  I didn't read that the selected pixels needed to be any particular color.   The pixels I selected were all white from an otherwise transparent layer.  I will try again with the white pixels turned black and the transparent pixels white, while I wait for another reply   Thank you.
