When I try to apply the lip sync, it will not read the audio file I put on the stage. I've tried both as a WAV and MP3, but the 'sync with audio in layer' feature is still grayed out.

Yes, exactly. The issue arises when trying to link the sync to the audio, the option is grayed out and states that there is no audio file even though it was imported to the stage.

you have an as3 document, not html5/canvas, correct?
if so, copy and paste here a screenshot showing your timeline sound and the properties panel showing the sound's sync property.

Note that both your audio and your nested mouth symbol have to be on the same "timeline level". For example, if your audio is on the main timeline and you try to use auto-lip sync while inside a graphic symbol's timeline the option will be grayed out; the feature can only "see" audio on the timeline you're currently at. To fix this, make a copy of the audio layer and paste it inside the symbol; don't worry, your audio won't be doubled, Animate mutes all nested sounds by default.
