Your question is very confusing. The file you're showing appears to be a native Illustrator file which would have the suffix .ai. The video you provided shows someone editing an Illustrator file. Please be specific as to what you are looking for.

I mean originaly what type of image extension file is used to trace. For example I took this in PNG screen shot & trace it.

Image trace is a feature within Illustrator. Its function is to make a vector line drawing from a raster image. The file then would be saved as an .ai file in order to keep it editable in the future. See https://helpx.adobe.com/illustrator/using/image-trace.html for more details.
