Pretty much as you see in the title.
It's a simple enough solution for me to workout, I just close Microsoft To Do... but it would be nice if I was able to keep it open whilst using one of the most important shortcuts/hot keys in Illustrator.

Thanks you so much for sharing this information. This issue has been reported by a few other users as well and in most of the cases it was found that there is some application which is conflicting with shortcuts. Does "Microsoft To Do" has some default set of keyboard shortcuts?

Hello @caseym67414055,
We understand that encountering technical issues can be frustrating. As suggested by Monika, other apps can interfere with keyboard shortcuts in Illustrator. Would you mind trying to run Illustrator under Safe Mode (Windows / macOS) and checking if it helps? If it works, you might need to manually find and disable/remove the conflicting app when using Illustrator.
