This could probably be performed with an Action or a Script – if the expectation is realistic for working one image at a time.

Thanks... From the same sample images that you provided, can you also provide the final 4000x4000px finished example as a reference?
Is it only the iris that needs to be a circle, or is it the pupil as well? You only mentioned the iris, but your example did have two regions shown with the pupil separate. It is easier if only the iris and then the pupil will just distort in proportion. But if you need both to be perfect circles then more steps are required.

I have included the 4000x4000px finished example from sample image of the blue eye in the link below.
The pupil also needs to be a circle yes, I forgot to specify. And ideally, the pupil would be 970px in diameter in the final step.
Yes, I think it would require more steps. It would be awesome if you could help me in any way!

I tested a proof of concept Action for the iris, the pupil would be similar.
Should the pupil be mathematically centred on the centre of the iris, or just be resized from the centre in the same relative position, even if off centre?
I'm thinking a 2 step interactive approach, first the iris, then the pupil second.
The vector ellipses would be adjusted by the user, the rest would be automated.

Do you want the pupil covered by the 970px black circle? Or is it that just the pupil itself is resized to 970px round?
Edit: What am I missing in how you made the off-centre pupil centred? What manual steps were taken? Content aware fill? The pupil is nowhere near the centre in the original.

I made the off-centre pupil centered with the warp tool. However I am looking for a more automated approach, with vector elipses.

Centering and scaling the pupil and subsequently not only scaling but also distorting the iris might be a real problem.
Puppet Warp will probably not work out, creating a Displacement Map for the Filter Displace might be possible but seems daunting, …; @Stephen Marsh , what is your approach to this?

Yeah, good question – I am now having to re-evaluate things as this wasn't initially obvious (but should have been). I'm hoping content aware fill will come to the rescue for automation... But this may mess up some scientific or medical use case which has not been stated.

The final use case is not for scientific or medical use, It is for aesthetic reasons. However, I would want to preserve as much of the original texture and color of the eye as possible 🙂
