    Always keep the layered PSD copy of your document (as well as a flattened one, such as a JPG, for distributing to others), so that you can always go back to your original image and make further changes, if necessary.

    Is the file unflattened now?

    If so, File > Save as > .psd

    If it is already flattened and hasn't been closed, try Edit > Step backward until you undo the flattening.

    As Peru Bob said .......and make sure "Layers" is checked in the "save as" dialogue (it normally is by default but can get unchecked if previous save as for example PDF)
