Open video file and Timeline. Go to Layer > New > Layer. New blank layer should be above video layer, if its not post screenshot of entire window including Timeline and Layers panel.

If yourr new blank layer is in the same video group you can drag it outside group then drag on timeline to position it as you want. Paint on new blank layer to achieve effect you are looking for.
