Glows and neon tube lights are just stacked duplicates of the same layer, blurred, recolored and mixed with blending modes like Add or Screen while tweking colors and opacities. The only bit of extra finesse in the sample image is of course the illusion of 3D by stacking the layers and in addition there appear to be a few subtle paint-over effects to give the illusion of a light sheen. the rest is just a matter of experimenting around and finding "nice" settings for everything.

I will try to give you basic instructions to get idea how that deed can be done:

  • Select two leaves from background with many leaves. I mean leaves which appear to be in front of neon stroke and duplicate with selection active so you have them on separate layer. You can experiment with Outer Glow layer effect to get some glow aroung leaves edges.
  • Craete rectangle or shape layer in between background layer with many leaves and two leaves on separate layer. Play with Layer effects like Outer Glo
