Define signature field using API only

I have a situation where all of the PDFs we need signed are created for each customer individually and on the fly.

I need to be able to define the signature field using the API only.

Is this possible? I cannot edit the pdf and add any sort of tags. It would have to be done using an x,y coordinate value.
