Brush work not visible on Paint on Transparent

Hey guys! I'm doing a scribble animation for a video and I want to add the glow effects to my brush work. However when I select Paint on Transparent, the Layer is still showing me pieces of the Artwork and not the brush work itself. I don't know what else to do. Please see pictures attached
