Can I make a camera effect on only one layer?

So that a 3D layer beneath isn't affected by the camera and I can use a different camera for it?

I don't think it is possible in a single composition. You'll need to use multiple compositions.

Precomp your 3D layer that you do not want to be affected by the camera in the main comp. Keep this Precomp as a 2D layer. You can create a camera inside the Precomp. I understand it's not the best solution but it should work.
