Does or can After Effects take advantage of two video cards?

The simple answer most definitely is "no", the more complicated answer could be "maybe". It all depends on the specifics of your GPU setup, what effects you use and so on just as well as what GPU-driuven third-party effects you may use. Generally, though, a vanilla AE simply won't be using more than a specific sub-set of GPU features, so investing in beefy GPUs is pretty much a waste of money.
