I have CS5 on my PC and I want to upgrade to CS6.
I would like to install CS6 on my wifes new Mac at the same time.
Can I purchase an upgrade that includes multiple platform capability?
I have only one installation activated at this time.

Only if you join the Creative Cloud.
One membership gives you up to two multi-platform (Mac and/or Windows) activations. So you can then activate Mac/Mac or Win/Mac or Win/Win.
To comply with the EULA (End User License Agreement) those two activations should be used by the same person and the apps must not be used at the same time. But I'm sure plenty of people share the two activations between themselves anyway.
If you stick with perpetual licenses (like you currently have), they are licensed per operating system. Then you will have to purchase a CS6 Windows upgrade (2 Windows only activations) plus a new Mac only full version of CS6 (2 Mac only activations).
