changing external drives

My source files are located in three external SSD drives all connected to the desktop via USB and each with a separate drive letter on the desktop. . I am in the process of transferring all folders from the three to one 4 TB SSD that in turn will be plugged via USB to the desktop with a drive letter than may be one of the three. Any suggestions on how to locate the new drive/folders globally?
