I have a mov file with a transparent background downloaded from the Internet (shown in photo number 1
Its size is only 54 MB. But when I export the same file again in MOV format, its final size is already 373 MB.
The only difference between them you can see in the picture (I marked them in red)
How can I export it with the same quality and the same original size of 54 MB

The Quicktime PNG codec was dropped by Apple several years ago (with the release of macOS Catalina which dropped 32-bit support). Adobe has included a read-only codec so you can work with it but the recomendation is to move towads ProRes4444 : Use Final Cut Pro to transcode Quicktime movie files that contain PNG frames - Apple Support
You won't be able to reencode to PNG video natively unless you have a machine still running a previous macOS version. If you have Quicktime Player for Windows you can import a render and export to legacy codecs (like PNG) if you have Quicktime Pro.
