I already had Adobe audition 3 (software) installed on my tower.  I upgraded to windows 10 and while  Audition opens and I can open files, the play function does not work.  HELP!

Audition 3 and Windows 10 really don't get on very well, I'm afraid. What's happened in your particular case is that the software isn't recognising your sound device - which is why the transport won't work. In theory, you go to Edit>Audio Hardware Setup and change the driver for the appropriate one for your sound device. I say in theory, because if I try this on this Windows 10 laptop with Audition 3, it crashes the OS to a BSOD. That's not supposed to happen at all on Windows 10, so whatever it is that it doesn't like, it's pretty serious. Caveat Emptor.

Please see previous reply - effectively we are unable to support Audition 3 on Windows 10. But you have told us nothing about your setup, so we can't advise further anyway, I'm afraid.

In Audition go to Edit - Audio Hardware setup -  for each tab (edit / multitrack) hit the Control Panel button and in the resulting window check your Headphones in the output list and move them up the list and hit OK. On the first panel then hit Apply. When both tabs doen hit OK. Worked for me!
