I would like to track movement and audio from a video file to animate a character. Is it possible?

Extract the audio from your video file into wav format (I don't remember if MP3 is acceptable or not) and import it into your scene with the puppet. Then select the Audio and click on  Timeline > Compute Lipsync from Scene Audio. The rest is automatic.
