I cannot open Encore CS6 Trial either with or without installing update EncoreTrial-6.0.1-mul-AdobeUpdate

I keep getting the following error -

"Adobe Encore CS6 cannot run in non-royalty serialized mode. The application needs to be serialized with a royalty bearing serial number."

As of my knowledge, Encore is one of the programs that do not run in trial mode, because of third party patent rights. You can run encore only, when having a licensed copy of Adobe products including the Encore program. This is not Adobe's choice.

No big deal to uninstall, but would have been helpful if Adobe had not included Encore in CS6 trial.

Yes. But if you buy the license, you will be able to use encore without reinstalling. Premiere is also "crippled" as some codecs are not working. But at least you can try the software!

Due to 3rd party license issues there is no such thing as an Encore trial

You MUST purchase Premiere Pro to then install and use the bundled Encore - Link to buy CS6 via telephone http://www.adobe.com/products/cs6.html

Or you may install PProCS6 if you have a Cloud subscription
