Is it possible to change or paint a brush color when a gradient is on the stroke ?
You can't apply a gradient to an art brush.
I remember you have asked this before.
You can build some appearance stack to get it working somehow, but it might be limited in a way that it doesn't do what you want to achieve.
And you might want to tell us what exactly you want to achieve. This small screenshot is not useful at all.

Can you please show something? Because as always it depends on how exactly the gradient has been applied and what exactly you'd like to achieve.

THose are bristle brushes, right?
No problem to apply a gradient to them. Actually that's pretty straightforward, just do it.

Regardless if I asked this question prior and if I did a link would help as I may want to reference that question. Otherwise, you mention you can't apply a gradient to an art brush, but you applied a gradient to an art brush live ?

That's a hack what I showed there. It may or may not apply to your situation.
Please explain in detail what you want to do and show the complete piece. Also please tell us everything about how this will be produced.

I applied Outline stroke effect to the path, then grouped it.
Applied pathfinder trim effect to the group, then a gradient fill.
