Red color one & black one is two line strokes. Drawn by using line tool only. One shows Expand & other shows Expand Appearance. What is the reason for that?

Generally, expand appearance is when something else (like a warp for example) has been applied and is live and therefore editable. It would also appear if an appearance had been added via the appearance panel. The video is blurry for me, so I am not sure if you did any of those things, but that is when expand appearance versus expand would show up.

Expand Appearance is there when you have used special appearances, effects, transparency, brushes ...

Above one was I simplified the main issue.
Real issue is I replicated this work but in my file Expand is grayed out only Expand Appearance is active. What is the reason for that?

It is still blurry, but as I explained, there is a difference between the two based on what you did to the object. As a practical matter, once you expand, the live effect will not be editable anymore. But you end with an expanded object in both cases.
