What is the best way to calculate ink coverage for a spot UV? We are only allowed 25% coverage on a front cover and need to work out how much is currently being used!

create a new swatch with 25% tint. Use this swatch.
If you have some overprinted inks. open the Separations Preview panel in Inc Limit moe and set the limit as 25%. The red areas will show the over limited states.

Unless the spot coverage is geometrical shapes, there is really no easy way to calculate area. I can't imagine if the spot UV was slightly over 25% that it would be an issue. Can you post an image of the spot plate, I'll gladly give a visual estimate of coverage.

Not sure why you are limited to just 25%, however...
You could place the cover as PDF and create a rough path of the UV area, or simply copy the path denoting the UV area from ID to AI.

Select the path and run the script in AI--it will give you the shape area in sq. inches.
Calculate percentage of shape vs page-size sq. inches.
